We have previously shown that FK-506 (tacrolimus) improves random-pattern skin flap
survival in normal animals. 1 Skin flap survival is decreased in cigarette smokers,
presumably by perturbing endothelial function and tissue viability. Nicotine, as the mediator
of this effect, impairs endothelium-dependent relaxation, which is thought to result from the
reduction in nitric oxide (NO) production and eNOS mRNA expression in exposed vessels. 2
